It's super inconvenient and primitive. You fill out some forms on BitInstant that basically gives them the info to prepare to look out for your transfer. Can be done @CVS, CHase, a few others. Then you have to go to CVS and there's some goofy phone you use and more forms to fill out there. Apparently it is very inconsistent store to store about how nosy the employee will be (Makes sense). You can lie about your info. However; you will be on security cameras. If you're doing over $500 it would be sketchy as fuck. It will require you to withdraw money from an ATM (likely fee there unless you are lucky enough to have free ATM withdrawals). BitInstant then charges I think 4%. At the end of the day if you withdraw $400 you would probably get 20 BTC from BitInstant. At this point, your bitcoins are "instant."
